The Monster was the first piece of mythology introduced on the show. It first appears during the night after the crash when the survivors hear a loud, unidentifiable sound coming from the jungle and witness trees being torn down in the distance. The next day, Jack, Kate, and Charlie see the power of the monster first-hand when it rips the pilot from the cockpit and leaves his mangled body in a tree. In "Walkabout", Locke also has a direct encounter but is spared. He later tells Jack, "I looked into the eye of this island, and what I saw was beautiful." In "Exodus: Part 2", Locke's second encounter provides the first on-screen glimpse of the monster (it appears to be an animate tendril of black smoke accompanied by strange mechanical-like sounds). In "The 23rd Psalm," Charlie and Eko have a confrontation similar to Locke's. As Eko stares down the monster, the black smoke briefly flashes images of Eko's past. It is worth noting that there are theories of the monster appearing in a different form. This includes the hallucination of Jack's father (wearing black) and the horse both Kate and Sawyer see (also black). In fact, the May 26 podcast of Lost claimed that viewers have seen the monster after The 23rd Psalm without realizing they were looking at it.

Prior to their arrival on the island, both major and minor characters had occasion to interact, often unknowingly, sometimes affecting each others' lives. These are revealed through characters' flashbacks, and are typically only obvious to viewers. An example occurs in the episode "Outlaws", when Sawyer meets a downtrodden doctor in an Australian bar, who turns out to be Jack's father, Christian. He reveals to Sawyer his inability to tell his son how proud he is of him. Later, on the island, Sawyer realizes the relationship between the two men, and shares with Jack his father's words. In another crossover, Ana-Lucia goes to Australia with Christian Shephard to act as his bodyguard. A less direct example is Jack's future wife, Sarah, who is said to have caused the car crash that killed Shannon's father. Most often, characters are oblivious to such intersections, such as Hurley's fast food manager, Randy, becoming Locke's superior at a box company, which Hurley then owns after winning the lottery; Sayid sharing a military transport with Kate's father; Sawyer being brought through a police station where Boone is being questioned; Sayid being dropped off from a truck by the man who shared the hatch with Desmond; Locke inspecting a home for Sayid's childhood friend, Nadia; Sawyer being waited on by Kate's mother in a restaurant; Claire's psychic admitting to Eko that he is a fake. Other crossovers are more fleeting, with characters appearing on televisions or being glimpsed in the background, such as Jin being at a house where a child was watching Hurley's lottery interview, or Jack passing Shannon as she is being told of her father's death in the hospital. Damon Lindelof has stated that these are not "Easter eggs", but rather a larger part of the mythology of the series.

"The Others" are mysterious inhabitants of the island whose presence predates the crash of Flight 815, as well as Danielle Rousseau's arrival. In addition to kidnapping Rousseau's daughter, Alex, as an infant, the Others have infiltrated the Flight 815 survivors' camps on at least two occasions, and are responsible for the deaths of at least two castaways and the unexplained disappearances of many others.

In the episode "Raised by Another," Claire was kidnapped by one of the Others, a man calling himself "Ethan Rom," and held against her will for a period of several days. She later returned in the episode "Homecoming," but with no memory of anything that had happened to her since before the crash. She eventually regained some of those memories in the 2nd season episode "Maternity Leave," in which it was shown that Ethan had kept her drugged throughout most of her captivity, as well as repeatedly injecting her with a substance apparently intended to help immunize her baby against some sort of illness he claimed was present on the island. Claire later escaped with the help of Alex Rousseau, and Ethan was killed by Charlie while trying to retrieve her.

At the end of the first season, the Others kidnapped Walt, shot Sawyer, blew up the raft that several castaways had built in an effort to escape the island, and left Michael, Sawyer, and Jin stranded more than 20 miles out to sea. During the second season, they abducted twelve tail-section survivors (including several children) and murdered at least one other. Rousseau later catches a man who claims to be "Henry Gale from Minnesota," who turns out to be some sort of power figure in the group.

The numbers 4, 8, 15, 16, 23 and 42 appear throughout the series, both in sequence and individually. They were broadcast from the Island's radio transmitter, and it was this message that drew Rousseau's expedition there. Although she later changes the message after the deaths of the rest of her team, the digits had also been heard by other people, eventually making their way to Hurley, who used them to win a lottery. After those around him suffer a series of misfortunes, he begins to believe the numbers are cursed. His search for their origin leads him to Australia. On the island, Hurley discovers the digits engraved on the hatch. They also appear inside the bunker, on medicine bottles, and constitute a code that must be entered into the Swan station's computer. The sum of these numbers, 108, has become significant in connection to the DHARMA Initiative.

According to the DHARMA Orientation video in the Lost Experience, the Numbers represent the core factors of the Valenzetti Equation.

The existence of the DHARMA Initiative is established by the film that Jack and Locke find in the Swan Station. It was founded in 1970 by University of Michigan doctoral candidates Gerald and Karen de Groot and financed through Alvar Hanso and his Hanso Foundation. It comprises a group of "scientists and free thinkers" from around the world who were brought together at a "large-scale communal research compound" to conduct research into various disciplines, including meteorology, psychology, parapsychology, sociology, zoology, and electromagnetism. According to the Swan's orientation film, the DHARMA Initiative has placed a number of research stations around the island. Four have been featured in the series thus far. The Swan station, commonly called "the hatch," is being occupied by the survivors. As part of the Lost Experience, DHARMA has been revealed as an acronym for "Department of Heuristics And Research on Material Applications."

The "Lost Experience" has recently revealed that the objective of the DHARMA Initiative is to alter 6 factors of the Valenzetti Equation revealed to have a huge impact on the date the human race will destroy itself, whether by global warming, chemical warfare, overpopulation or many other possible methods. These factors are represented as numbers in the Valenzetti Equation and are also the numbers frequently mentioned in the show: 4, 8, 15, 16, 23 and 42.

There have been a number of instances in which the survivors encounter animals that are either not native to the island or have special attributes. In "Pilot: Part 2", Sawyer shoots a charging polar bear, which cannot naturally survive in a tropical environment. In the third season's first episode, Sawyer is told that "it took only four hours for the bears" to find out how the mechanism works in the cage where Sawyer is held. The mechanism gives food and it gives a picture that bears have been kept in that cage. Later in the first season, Walt has an encounter with the same species when he is attacked after wandering into the jungle. In "Outlaws" Sawyer has several run-ins with a boar that he believes is purposely harassing him. Kate and Sawyer see a black horse in "What Kate Did" and flashbacks in that episode reveal that Kate believes she has seen it previously.
